This experiment was conducted to investigate the effect of growth regulators (BA, NAA), sucrose, MS inorganic salts (MS), and activated charcoal (AC) on the growth of each stage in shoot tip culture of apple rootstock M.26.
The results were summarized as follows:
1. In culture establishment stage, only BA at an optimum of 2.0§·/§¤, not NAA which produced callus, was required for shoot growth.
2. In shoot multiplication stage, both shoot multiplication and growth were achieved with the combination of 2.0§·/§¤ BA and 0.5§·/§¤ NAA, and 30g/§¤ sucrose was also effective.
3. In rooting stage, the best rooting was obtained with 0.1§·/§¤ NAA. AC showed inhibition in rooting percentage and root number, except root length. Both 30g/§¤ sucrose and 1/4MS were effective for rooting.
